Leica EMGP

The EMGP is an automatic plunge freezer device. The sample is diposited on an electron microscope grid followed by blottind. The thin sample layer is vitrified by rapid freezing into liquid ethane.

Characteristics

  • Chamber temperature (+4°C - 60°C)
  • Chamber humidity
  • Blotting time
  • Ethane temperature

Sample preparation

  • 2 grids per samples with 2 different blotting times
  • Different filter papers can be used depending on sample viscosity
  • 10 nm gold nanoparticles (Duchesne et al., 2008) added for cryo electron tomography

Requirements

  • No hazardous materiel
  • High concentration (108 - 1012 particles/ml)
  • Low concentration of "matching" compounds such as sucrose or glycerol
